Abstract
The PhenX (consensus measures for Phenotypes and eXposures) Toolkit (https://www.phenxtoolkit.org/) is a publicly available knowledgebase of well-established, broadly validated measurement protocols recommended for use in genomic, clinical, translational, and epidemiological research involving human participants. Consistent use of PhenX protocols, including questionnaires, bioassays, and physical assessments, improves the rigor, transparency, reproducibility, and comparability of research data. The Toolkit was recently highlighted in the National Institutes of Health (NIH) implementation plan, "Leading in Gold Standard Science," as an example of efforts to advance reproducible biomedical research. The Toolkit currently includes 1010 protocols spanning a wide range of diseases, exposures, and study designs. Content development is guided by the scientific community through a consensus-based process involving expert Working Groups, community input, and strategic oversight from the PhenX Steering Committee. Collaborations with NIH Institutes have expanded Toolkit content in priority areas, including substance use and cardiovascular disease. The PhenX Toolkit enhances study design and supports implementation of selected protocols. PhenX also collaborates with resources that facilitate data collection (REDCap), data standards (LOINC), and data repositories to promote interoperability and cross-study analyses. To date, the Toolkit has been recommended in 711 NIH funding opportunities and cited in >1300 publications. Future directions are discussed.
