Abstract
Background: Eosinophils are viable for a limited period of time if they do not receive external stimulus. We hypothesized that eosinophils direct contact via integrins is important factor for their viability and effect for development of airway remodeling in asthma, especially after allergen provoked acute asthma episodes. Aim: To investigate adhesion of eosinophils to airway smooth muscle cells and its effect on their viability in asthma. Methods: We used individual combined cell cultures of hTERT airway smooth muscle (ASM) cells and blood eosinophils. Eosinophils were isolated by high density centrifugation and magnetic separation from 12 allergic asthma patients and 11 healthy individuals before and 24h after bronchial allergen challenge (BAC) with inhaled home dust mite allergen. Eosinophils adhesion was evaluated by measuring eosinophil peroxidase activity; eosinophils viability was evaluated with flow cytometer after Annexin V-PI staining. Data presented as mean±SEM. Results: After 1 h 69.5±5.4% of added asthmatic eosinophils adhere on ASM cells, while adhesion of healthy eosinophils was 49±4.1% (p<0.05). Moreover, after BAC asthmatic eosinophils showed 17.3±6.8 % (p<0.05) increased adhesion, while it has no effect on healthy eosinophils. Furthermore, after 24 h of incubation with ASM cells, non-viable asthmatic eosinophils number decreased by 42.3±4.0 %, healthy eosinophils by 22.6±2.5 % (p<0.05), comparing with eosinophils without co-culturing with ASM cells. After BAC non-viable asthmatic eosinophils number decreased by 53.3±6.4%, comparing with eosinophils before BAC (p<0.05). Conclusion: Increased eosinophils adhesion to ASM cells enhanced their viability in acute allergic asthma.
