Abstract
Bu çalışmada, koliform grup bakteriler/fekal koliform bakteriler/E. coli’nin EMS yöntemi ile sayımında kullanılan TS/ISO 6063 yönteminde analiz süresinin kısaltılabilme olasılığı araştırılmıştır. Denemelerde EC Broth besiyerine %0.1 triptofan ilavesi ile kombine edilmiş bir besiyerinde 290 adet bakteri 44.5±0.5 oC’da gelişme, gaz ve indol oluşturma bakımından analiz edilmiştir. Buna ilave olarak 290 bakteri için LST Broth+MUG besiyeri kontrol olarak kullanılmış, bu besiyerinin TS/ISO 6063’e alternatif olup olamayacağı incelenmiştir. Araştırma sonuçlarına göre EC Broth+Triptofan kombine besiyeri ile kontrol besiyeri olarak kullanılan LST Broth+MUG besiyeri E. coli için %100 uyum göstermiş ve buna göre 6 gün süren bir analiz yöntemi yerine EC Broth+Triptofan ile 3 gün süren analiz yönteminin kullanılabileceği, LST Broth+MUG besiyeri ile denenen 290 bakteri arasında sahte negatif ve sahte pozitif floresan reaksiyonu alınmadığı görülmüştür.
