Abstract
Objective: To evaluate the trypanocidal, leishmanicidal and cytotoxic activity of Eugenia jambolana (E. jambolana) and Eugenia uniflora (E. uniflora) extracts and fractions. Methods: The products were characterized by LC–MS. Antiparasitic assays were performed and cytotoxicity was evaluated in fibroblastos. In vitro assays were performed using spectrophotometric evaluation. All assays were performed in thrice. Results: The results showed that the extracts and the tannic fraction from E. jambolana inhibited 100% of the epimastigote lines. The ethanolic extract was the most efficient in all concentrations tested against the three parasite strains. In the cytotoxicity assay the flavonoid fraction showed low toxicity. All E. uniflora samples showed cytotoxicity at the highest concentration tested, but the extract showed no toxic effect on the fibroblasts at the lowest concentration. The flavonoid and tannic fractions were more efficient against Leishmania braziliensis promastigotes compared to the extract. However, the extracts and the tannic fraction were more effective against Leishmania infantum strains. The effect on epimastigote cells was observed at all concentrations tested, with all E. uniflora samples. However, the samples were more effective at the highest concentration, where there was inhibition in 100% of the Trypanosoma cruzi strains. Conclusions: The species E. jambolana and E. uniflora presented antiparasitic activity against all tested parasite strains, indicating that these species can serve as an alternative therapy as they were efficient in the tests performed. The E. uniflora extract and the E. jambolana flavonoid fraction presented a low cytotoxicity, opening the floor for new biological studies.
Zielsetzung: Evaluierung der trypanoziden, leishmaniziden und zytotoxischen Aktivität von Extrakten und Fraktionen aus Eugenia jambolana (E. jambolana) und Eugenia uniflora (E. uniflora). Methoden: Die Produkte wurden mittels LC-MS charakterisiert. Es wurden antiparasitäre Assays durchgeführt und die Zytotoxizität an Fibroblasten evaluiert. Die In-vitro-Assays erfolgten mittels spektrophotometrischer Auswertung. Alle Assays wurden in dreifacher Ausführung durchgeführt. Ergebnisse: Die Ergebnisse zeigten, dass die Extrakte und die Tanninfraktion aus E. jambolana 100 % der epimastigoten Linien inhibierten. Der ethanolische Extrakt war in allen getesteten Konzentrationen gegen die drei Parasitenstämme am wirksamsten. Im Zytotoxizitätsassay wies die Flavonoidfraktion eine geringe Toxizität auf. Alle E.-uniflora-Proben zeigten bei der höchsten getesteten Konzentration eine Zytotoxizität, der Extrakt zeigte jedoch bei der niedrigsten Konzentration keine toxische Wirkung auf die Fibroblasten. Die Flavonoid- und Tanninfraktionen waren im Vergleich zum Extrakt wirksamer gegen Promastigoten von Leishmania braziliensis. Die Extrakte und die Tanninfraktion waren jedoch wirksamer gegen Leishmania-infantum-Stämme. Die Wirkung auf epimastigote Zellen wurde bei allen getesteten Konzentrationen bei sämtlichen E.-uniflora-Proben beobachtet. Die Proben waren jedoch bei der höchsten Konzentration am wirksamsten, bei der eine Inhibition von 100 % der Trypanosoma-cruzi-Stämme vorlag. Schlussfolgerungen: Die Arten E. jambolana und E. uniflora wiesen eine antiparasitäre Aktivität gegen alle getesteten Parasitenstämme auf. Dies deutet darauf hin, dass diese Arten als alternative Therapieform dienen können, da sie sich in den durchgeführten Tests als wirksam erwiesen haben. Der E.-uniflora-Extrakt und die Flavonoidfraktion von E. jambolana wiesen eine geringe Zytotoxizität auf, was den Weg für neue biologische Studien ebnet.
